Hi, when I practice Siddhasana, at a moment in the meditation it’s became unconfortable, so I take off my heel and continue my practice of meditation with “I AM”, and I feel the tip of my penis extremly sensitive with tingling. What does that mean? Thank tou by advance

As Maheswari mentioned, this sensation doesn’t indicate anything concerning—there’s nothing to worry about, though it may feel unusual at first.
When the root chakra is stimulated, increased prana flow to the genital area commonly occurs, which explains the tingling sensation you’re experiencing. Sometimes the opposite can happen, where the area may feel numb instead. There’s actually a dedicated lesson covering this phenomenon called “Sleeping Lingam.”
You’re absolutely right to modify your practice when experiencing discomfort. As your body adapts and your practice develops, you’ll eventually be able to hold siddhasana comfortably for extended periods.

When doing siddasana you have to be careful not to put any pressure anywhere near the sexual organs. That is very important, it can cause various effects from mild to severe.
A very light touch, barely noticeable is all you need. Hopefully what you felt will go away, give that a try and if it happens again adjust your position and monitor the effects.
If it keeps re occuring stop siddasana and find another posture.
But this condition should go away just by easing any pressure.

Yesterday, I discovered this sensation is associated with my concentration on the third eye and it is also present during the pranayama with the pressure of the heel, not only when I take it off during the meditation.

If reducing the pressure at the perineum during siddhasana helps to solve the issue, then that is great. If you still find that you get the increased sensitivity and tingling at the tip of the lingam when you do not use siddhasana, then you could simply be experiencing the activation of the energetic channel (nadi) inside the lingam. This is nothing to be concerned about and symptoms will fade over time as the nadi is purified.
